This kit is intended for quantification of double-stranded DNA in the broad range with fluorometer (any version can be used). The kit includes QuDye dsDNA BR fluorescent dye concentrate 200x, buffer for preparation of dye working solution, and DNA concentration standards (0 and 100 ng/µL). QuDye dsDNA BR Reagent is selective to double-stranded DNA, so measurement results are not affected by RNA, free nucleotides, or protein contaminants. Other minor contaminants such as salts, detergents, and solvents have a non-significant effect on measurements results. This kit allows accurate dsDNA quantification in the range from 100 pg/µL to 1000 ng/µL for initial sample (DNA amount range for measuring: 2–1000 ng of DNA in 200 µL of the test sample).

The main kit component is QuDye dsDNA BR fluorescent dye, which selectively interacts with DNA and exhibits enhanced fluorescence upon binding to DNA molecule. This kit allows convenient dsDNA quantification in the broad concentration range and does not require a large volume of the initial sample (1–20 µL is required for the assay). All measurements are performed at room temperature and take on average 15–20 minutes (depending on the sample amount). Sample fluorescence is stable for 3 hours. To perform a measurement, prepare dye working solution, dilute DNA standards and experimental samples with it in individual tubes and perform fluorescence measurements using fluorometer. For your convenience, we offer kits including thin-wall tubes compatible with fluorometer.

This kit can be also used with QuDye Protein Quantification Kit to determine protein contaminants in the DNA sample.

For quantification of dsDNA of lower concentrations, use QuDye dsDNA HS Assay Kit with the range of initial sample concentrations of 10 pg/µL to 100 ng/µL.
