ROX (5-carboxy-X-rhodamine) is a passive reference dye for qPCR application. ROX is used for fluorescence signal normalization; its signal depends on neither amplification reaction nor the template amount in the reaction. Use of the dye allows reducing well-to-well variability caused by differences in reaction volumes due to pipetting errors, variations in optical path length, presence of air bubbles in the wells.

A lot of real-time PCR systems* require ROX as an internal reference dye for signal normalization. To normalize the fluorescence intensity data, the fluorescence intensity of the reporter dye (for instance, dsGreen) is divided by the fluorescence intensity of the ROX passive reference dye; this ratio is known as the normalized reporter intensity (Rn).
