Click-monosaccharides for metabolic labeling of glycans

Most modern biological studies use methods that specifically identify molecules of interest for visualization, isolation, or quantification. Hybridization methods are used to detect nucleic acids. Proteins are monitored using specific antibodies or labeling. However, none of these methods are suitable for glycan detection. For a long time, the only tools for their detection were lectins and antibodies—methods that have limitations when working with glycans related to their specificity and affinity.

Click chemistry helped overcome the problem of specific labeling of glycans. Carolyn Bertozzi proposed introducing bioorthogonal groups into sugar residues of interest and, after they are metabolized by the body or cell, identifying them through click chemistry reactions. The importance of this method was recognized by the Nobel Prize in Chemistry in 2022.
