The DSSO (Disuccinimidyl sulfoxide) linker is an MS-cleavable, homobifunctional crosslinker engineered for modern cross-linking mass spectrometry (XL-MS), used for high-resolution analysis of protein-protein interactions (PPIs) and structural topology.

The unique feature is a central sulfoxide group that undergoes predictable, collision-induced dissociation (CID) during tandem MS (MS/MS). This cleavage yields characteristic doublet peaks in the MS/MS spectrum. This distinct fragmentation signature enables unambiguous identification of cross-linked peptides and facilitates automated data processing, directly addressing the complexity of proteome-wide cross-linking studies.

DSSO provides robust, lysine-specific covalent stabilization of protein complexes, delivering precise, residue-level constraints for integrative structural modeling.
